Hi
I am new to Mister and was wondering if I can use the second SD card slot to hold an SD card with roms or if the roms need to be placed on the same SD card as the software (i.e. the 1st SD card slot)

Hi there,
The second SD card is directly wired to the FPGA and is generally used as raw storage on some cores. You wouldn't be able to access any ROM files from it.
You'd be better with a USB key or USB SSD (or USB HDD if you have a 5 volt power supply capable of 4 amps or more) to store your roms. MiSTer will even detect USB ROM storage and default to it.
